Emerging Treatment Approaches for Parkinson’s disease

Parkinson’s disease (PD) is the second most common neurodegenerative disease, manifesting as a characteristic movement disorder with a number of additional non-motor features. The pathological hallmark of PD is the presence of intra-neuronal aggregates of α-synuclein (Lewy bodies). The movement disorder of PD occurs largely due to loss of dopaminergic neurons of the substantia nigra, resulting in striatal dopamine depletion. There are currently no proven disease modifying treatments for PD, with management options consisting mainly of dopaminergic drugs, and in a limited number of patients, deep brain stimulation. Long-term use of established dopaminergic therapies for PD results in significant adverse effects, and there is therefore a requirement to develop better means of restoring striatal dopamine, as well as treatments that are able to slow progression of the disease. A number of exciting treatments have yielded promising results in pre-clinical and early clinical trials, and it now seems likely that the landscape for the management of PD will change dramatically in the short to medium term future. Here, we discuss the promising regenerative cell-based and gene therapies, designed to treat the dopaminergic aspects of PD whilst limiting adverse effects, as well as novel approaches to reducing α-synuclein pathology. Treatment of Parkinson’s disease through Personalized Medicine and Induced Pluripotent Stem Cells

Parkinson’s disease (PD) is an intractable disease resulting in localized neurodegeneration of dopaminergic neurons of the substantia nigra pars compacta. Many current therapies of PD can only address the symptoms and not the underlying neurodegeneration of PD. To better understand the pathophysiological condition, researchers continue to seek models that mirror PD’s phenotypic manifestations as closely as possible. Recent advances in the field of cellular reprogramming and personalized medicine now allow for previously unattainable cell therapies and patient-specific modeling of PD using induced pluripotent stem cells (iPSCs). iPSCs can be selectively differentiated into a dopaminergic neuron fate naturally susceptible to neurodegeneration. In iPSC models, unlike other artificially-induced models, endogenous cellular machinery and transcriptional feedback are preserved, a fundamental step in accurately modeling this genetically complex disease. In addition to accurately modeling PD, iPSC lines can also be established with specific genetic risk factors to assess genetic sub-populations’ differing response to treatment. iPS cell lines can then be genetically corrected and subsequently transplanted back into the patient in hopes of re-establishing function. Current techniques focus on iPSCs because they are patient-specific, thereby reducing the risk of immune rejection. The year 2018 marked history as the year that the first human trial for PD iPSC transplantation began in Japan. This form of cell therapy has shown promising results in other model organisms and is currently one of our best options in slowing or even halting the progression of PD. Here, we examine the genetic contributions that have reshaped our understanding of PD, as well as the advantages and applications of iPSCs for modeling disease and personalized therapies. Introduction About the Topic

Parkinson’s disease is a disorder of movement. The main system that is affected by this disorder is the nervous system. Parkinson’s disease is a chronic disease because the symptoms gets worse over time.

There are many other disorders that affects one’s movement. These include ataxia, cerebral palsy, and of course, Tourette syndrome. These disorders occur when an individual’s nervous system undergoes a change. By this, the person’s ability to stay still or to move becomes affected.

A study by the National Institutes of Health (NIH) has shown that at least 50,000 persons are diagnosed of Parkinson’s disease in the United States every year. What’s more? At least 500,000 persons are already living with the condition.

It is important to note that the symptoms of Parkinson’s disease do not occur all at once. As a matter of fact, they have a gradual development. The first symptom is often a slight tremor and a stiff feeling in the body.

With time, other symptoms show up. Some patients do have dementia.

Majority of the symptoms of Parkinson’s disease are caused by a reduction in the levels of dopamine. Dopamine is produced in the brain.

A 2015 study conducted in France has shown that men are kore at risk of developing Parkinson’s disease compared to the female gender. However, women have a higher risk as they get older.

In many patients. The first symptoms show up from 60 years of age or above. Studies have shown that in 5-10% of cases, the symptoms came up much earlier than usual. This is referred to as early-onset PD.

How do i know that i have parkinson’s disease? What are the symptoms?

The early symptoms of Parkinson’s disease include:

  • Tremor in the hands
  • An inability to coordinate or balance oneself. The patient cannot hold an object steadily. More often than not, it falls out of the patient’s hands.
  • A change in posture or gait. The affected patient tends to lean forward slightly, as if they are in a hurry.
  • The facial expression changes and assumes a kind of fixed state. This is attributed to the changes that control the muscles of the face.
  • The voice also undergoes a change. You will notice a tremor in the voice, or it may become softer than usual.
  • A small or cramped handwriting
  • A distorted sense of smell and issues with sleep.

Other symptoms that are typical of Parkinson’s disease include:

  • skin problems
  • constipation
  • sleep problems
  • difficulty in urinating
  • Problems with chewing or swallowing.

What actually causes parkinson’s disease?

The actual cause of Parkinson’s disease is not fully understood. However, the disease comes up when the brain cells die off.

Pathological conditions that may contribute to the onset of Parkinson’s disease include:

  • Reduced dopamine levels
  • Low levels of norepinephrine
  • The presence of Lewy bodies
  • Genetic factors and autoimmune factors

Current Treatment at Other Clinics

We understand that most features of Parkinson’s disease are caused by death of dopamine-secreting neurons. Thus, current treatment is channeled at replacing dopamine or administering drugs that mimic its functions. Such drugs include dopamine agonists or L-dopa. The bad news is that these drugs do have their own side effects which include dyskinesias (involuntary movement). It also causes neuropsychiatric problems. Thus, the drugs may ultimately become a problem to the individual.

As the disease gets more complicated, surgical stimulation of the brain tissue may be used. However, this only offers temporary relief.

We are different from many other clinics who offer stem cell or PRP.   Other may also not offer option of bone marrow derived stem cell, Adipose derived stem cell & umbilical cord stem cell.  They in effect expose patients to more invasive treatments like drilling bones without offering other easier options.  They also may use xray with unnecessary radiation for injections that can be easily avoided in most cases by using ultrasound.  Furthermore, they may not have proper knowledge of how other modalities can enhance the effects of Stem Cells.  These other modalities are used by us to focus on the whole joint and treating different parts of it instead of just treating one out of several diseased structures in the joint.

Safe Non Surgical Option

We use a combination of newest cutting edge technology in regenerative medicine to stimulate body’s own repair ability to reverse aging and damage.   We use Stem Cells as part of treatment plan designed by the doctor to treat the body.    These are safe investigational treatments that have shown very promising & safe results in thousands of treatments over the 10-20 years in Europe and USA.   They don’t involve using any medications or drugs.

What Does Stem Cell Therapy Do?

The researchers believe that the therapy works by:

Releasing cytokines into the body to prevent further damage and inflammation.

Preventing inflammation that can make the disease worse.

Creating healthy new tissues or missing or damaged products in body.

Many patients experience improvements in the treated area and other areas, like in a Mayo Clinic study where patients benefited in both knees after experiencing injections in only one.

How does stem cell therapy help with the treatment of parkinson’s disease?

Stem cells help in the treatment of Parkinson’s disease. Current research has assisted scientists to discover ways by which this disease is triggered and how they could be treated.

Research has shown that replacement of dead dopamine-secreting neurons in the brain can help to improve the condition of PD patients. Implanting dopamine-secreting cells at the affected sites gives a clinical response that is the same as that given by dopamine drugs. The advantage here is that the stem cell generated neurons will release the dopamine at physiological levels, at the site where it is needed, thus helping to curb the side effects that are typical of the other methods of treatment.

What makes our facility special?

At our Los Angeles facility, we have invented modern approaches to medical care. Our approach to treatment is second to none. The harvesting process does not involve any form of drilling, and the stem cells are purified and tested for quality before injection. We also employ ultrasound to help in the precise location of high dose stem cells at the required site. These cells will then perform their duty, promoting the formation of new tissues. We are experts in this and has seen positive outcome with our treatments. Our licensed physicians have undergone intense training running into thousands of hours, and so, you can be sure that you are in safe hands.

Frequently Asked Questions

What is Parkinson’s disease?

Parkinson’s disease is basically a neurological disorder. James Parkinson gave the first description for this disease in 1817.

What are the symptoms of Parkinson’s disease?

Common symptoms of this disease include tremor even at rest, bradykinesia, poor posture, and rigidity. For a person to be diagnosed, he must present with at least two of these four symptoms.

How is the disease diagnosed?

Diagnosis of Parkinson’s disease is different from that of other disease. There is no need for blood tests. The clinician observe the patient’s physical symptoms and then makes his or her diagnosis based on that. However, one of the conditions to receive a diagnosis is manifesting two of the four physical symptoms listed above.

What are the current forms of treatment?

Medications are used, and they include:

-Carbidopa/Levodopa (Sinemet)
-Pramipexole (Mirapex)
-Pergolide (Permax)
-Ropinirole (Requip)
-Bromocriptine (Parlodel)
-Entacapone (Comtan)
-Amantadine (Symmetrel)
-Selegiline (Eldepryl)

Can stem cell therapy cure Parkinson’s disease?

This varies from patient to patient depending on factors such as the patient’s present state of health, medical history, the extent of damage, and a host of other factors. You should note that the use of allogenic stem cells for the treatment of Parkinson’s disease is maybe effective for this condition. However, patients should try not to create the false expectation of a guaranteed cure.

What type of stem cells are used for the treatment of Parkinson’s disease?

We offer mesenchymal stem cells obtained from umbilical cord tissue. This tissue has a high multiplying ability which gives our medical team the chance to get a large amount of stem cells to treat the condition.

Is stem cell therapy safe for Parkinson’s disease?

We are confident in our ability to effectively treat your condition. All stem cell treatments are thoroughly reviewed by well-trained physicians and specialists. We are very honest with our patients, and if we think that your condition cannot be treated, we will let you know.

From where do stem cells originate?

Stem cells are present in many tissues and organs. The bone marrow is the richest source of stem cells. However, the most convenient region for harvesting is the umbilical cord.

How are adult stem cells harvested?

The physician draws the stem cells from the umbilical cord. The stem cells and the growth factors will then be concentrated using specially designed equipment. The physician then injects the cells into the destined regions.

What’s the Cost of Stem Cell Injection?

The cost of treatment depends on the procedure performed.  We often offer discounts for returning customers or larger packages of treatment using repeated injections or combining different treatments.  We also offer a price match policy on comparing treatments done by board certified physicians.  We also have interest free payment plan options including care credit for qualifying patients. A limited number of insurance companies and Workers compensation may at times cover some of the expenses.  You may be able to get reimbursed by your insurance for your doctor visits & ultrasound cost. The consultation fee can be deducted from your future procedures, making your consolation free of cost to you.

Book a consultation with our experts to see the type of procedure that is right for you.

When will my results show up?

Stem cell therapy relies on your own regenerative healing to occur. We are all different and so is our healing process. Some patients may begin to see their results within few weeks while others may require some months. The time taken for healing to manifest also depends on the severity of the disease.

What can stem cell therapy potentially treat?

Stem cell therapy is beneficial to a number of conditions such as osteoarthritis, erectile dysfunction, joint pain, renal disease, neurological disorders, autism, cellular rejuvenation, etc.

I’m finding it difficult to quit smoking. What can I do?

You can join a support group or employ the services of a therapist. Quitting smoking reduces your risk of contracting COPD by over 50 percent.

I’m on supplemental oxygen but I still have dyspnea. What do I do?

Low oxygen levels occur when the blood vessels do not connect with the air sacs. Because of this, the oxygen in the air sacs are not transferred to the blood and the individual becomes short of breath.

I’ve been diagnosed with COPD. How long can I live and what quality of life will I live?

Even with a COPD diagnosis, you can live as long and as healthily as you want. What you need do is to engage in safe exercise while also paying close attention to symptoms of acute exacerbation.

I’ve been hospitalized twice because of COPD. How can I avoid these in the future?

To remain stable and eliminate or decrease these episodes, you must learn about triggers, early warning signs of COPD flare-ups (exacerbations), and have a written action plan worked out with your health care provider. Taking your medications as prescribed will also help you stay healthy.
