Mesenchymal Stem Cell-Based Therapy for Kidney Disease: A Review of Clinical Evidence

Mesenchymal stem cells form a population of self-renewing, multipotent cells that can be isolated from several tissues. Multiple preclinical studies have demonstrated that the administration of exogenous MSC could prevent renal injury and could promote renal recovery through a series of complex mechanisms, in particular via immunomodulation of the immune system and release of paracrine factors and microvesicles. Due to their therapeutic potentials, MSC are being evaluated as a possible player in treatment of human kidney disease, and an increasing number of clinical trials to assess the safety, feasibility, and efficacy of MSC-based therapy in various kidney diseases have been proposed. In the present review, we will summarize the current knowledge on MSC infusion to treat acute kidney injury, chronic kidney disease, diabetic nephropathy, focal segmental glomerulosclerosis, systemic lupus erythematosus, and kidney transplantation. The data obtained from these clinical trials will provide further insight into safety, feasibility, and efficacy of MSC-based therapy in renal pathologies and allow the design of consensus protocol for clinical purpose. AN INTRODUCTION TO KIDNEY FAILURE

Everyone has a kidney. It is a very important organ of the urinary/excretory system. Normally, there are two kidneys in the human body. These kidneys are located towards the lower part of the back. Both kidneys are located on either side of the spine. Kidneys are responsible for the filtration and excretion of toxins. These toxins are sent to the bladder. The bladder then flushes out the toxins as urine.

When kidneys fail in their excretory and filtration function, then one is said to have kidney failure. So many factors contribute to the deterioration, and loss of kidney function. These include:

  • Exposure to toxins and pollutants in the environment
  • Some chronic diseases and acute diseases such as diabetes, high blood pressure, lupus
  • Trauma to the kidney
  • Severe dehydration.

When the kidney fails in its function, the body becomes loaded with toxins. The end result is a failure of the kidney, which may be very fatal.

How do i know that i have kidney failure?

The following symptoms will be experienced by a person whose kidney has failed. In some cases, there may be no symptoms at all. Possible symptoms of kidney failure include:

  • A reduction in the amount of urine that is produced by the kidneys
  • Swelling of the ankles, legs, and feet. This is caused by retention of fluids due to the inability of the kidney to excrete them.
  • Dyspnea (shortness of breath)
  • Chronic fatigue or drowsiness
  • Nausea
  • Confusion
  • Pressure or pains in the chest
  • Coma
  • Seizures

What causes kidney failure?

Many conditions can contribute to failure of the kidney. The causative factor also determines the type of kidney failure that an individual suffers. People at high risk of kidney failure usually have any of the following causes:

Poor blood flow to the kidneys

Reduced blood supply to the kidneys or sudden interruption of blood to the kidneys can trigger a failure of this delicate organ. Some factors that may cause poor blood flow to the kidneys include:

  • Heart disease
  • Heart attack
  • Burns
  • Dehydration
  • Allergic reactions
  • Infections (such as sepsis)
  • Liver failure

Anti-inflammatory medications and high blood pressure may also contribute to this disorder.

Issues with elimination of urine

When you are unable to urinate, toxins will build up in your kidneys. Certain kinds of cancer can also interfere with the flow or urine. These include;

  • Colon cancer
  • Cancer of the prostate
  • Bladder cancer

Other factors that can cause a shortage in blood flow to the kidney include:

There is no cure for kidney disease and the only treatments are to slow progression of the disease. 

After renal failure there are two major forms of treatment – dialysis and kidney transplant. Dialysis involves the use of a machine to filter the blood and purify it. This machine does the job of the kidney. It should be noted that dialysis is not a cure for kidney. It only helps the kidney to do its job, and may extend your life if done regularly. A kidney transplant is another treatment option. With a kidney transplant, you will no longer require a dialysis.

The mesenchymal stem cells are derived from umbilical donated after normal, healthy births. The medical history of each mother is screened. Test for infectious diseases is done, and each family gives their consent before donating.

All stem cells derived from umbilical cords are screened for infectious ailments. Screening is done to International Blood Bank Standards prior to usage.

As a matter of fact, a very low percentage of donated umbilical cords scale through this screening process.

From where do stem cells originate?

How are adult stem cells harvested?
